dc.description.abstractEl cultivo del café ha sido tradicional en Centro América y ligado al desarrollo socio-económico, generó el 3,8 por ciento del Producto Interno Bruto en 1996 y ocupa el tercer lugar en la producción en el mundo. La producción se concentra mayormente en manos de pequeños agricultores, además es una fuente importante de generación de empleo. La especie más cultivada en el mundo es C. arabica originaria de las regiones altas de Etiopía y Kenia y cultivada en las tierras altas de América Tropical. El material genético actualmente cultivado tiene como origen una base genética muy estrecha, derivada básicamente de dos poblaciones de porte alto Typica y Bourbon a partir de muy pocas semillas introducidas en el Siglo XVIII y XIX de Holanda y la Isla Bourbon. Con el descubrimiento y selección de una planta de porte bajo en Brasil, se desarrolló la variedad Caturra, la cual permite cosechas más fáciles y densidades de siembra más altas, pero susceptibles a la roya del café (Hemileia vastatrix) y los nemátodos. Otro esquema de mejoramiento genético se generó a partir de los Híbridos de Timor, las variedades que derivan del Híbrido de Timor son resistentes a la roya, a algunas especies de nemátodos y a algunas cepas de la antracnosis del fruto (Colletotrichum kahawae). Los objetivos de esta investigación fueron evaluar en el CATIE, Turrialba, Costa Rica, el poder de los marcadores moleculares para la caracterización varietal de café y evaluar la diversidad genética del material cultivado (Typica, Bourbon, Silvestres, e Introgresados) de café por marcadores moleculares y observaciones agro-morfológicas. Para la caracterización agro-morfológica se utilizaron características de la planta, fruto y grano. Se determinó para las características morfológicas una alta variabilidad genética entre variedades y entre árboles. La variación entre variedades es producto de la variación genética, en tanto que la variación entre árboles es producto de la influencia del medio ambiente, factores fisiológicos y de manejo del germoplasma, por ser materiales homocigotos. El color del brote de café es una característica importante para clasificar las dos poblaciones básicas de Typica y Bourbon. Los componentes de varianza indican que el factor variedad es el que más contribuye a la variabilidad observada, y los índices de heredabilidad demuestran que es factible obtener progresos genéticos en programas de selección en las variedades estudiadas. Las características de fruto, grano y fertilidad permitieron determinar la variabilidad genética presente entre grupos de café. La característica de relleno del fruto se presentó como la más importante para clasificar variedades entre el grupo Typica y Bourbon. Para la caracterización molecular se utilizó RAPD. El polimorfismo detectado fue alto debido a la presencia de genotipos de C. canephora y al grupo de introgresados, sin embargo, el número de marcadores polimórficos fue bajo. Por medio del cálculo de las similitudes entre individuos, dendrogramas y análisis de componentes principales se estimó las distancias genéticas entre grupos, y se generó la clasificación de los grupos genéticos. Se identificó los marcadores diferentes entre cada grupo y los marcadores comunes que se comparten entre grupos. Se determinó que la distancia genética entre el grupo Typica y Bourbon fue no significativa, demostrando la estrecha base genética que caracteriza estos grupos; sin embargo la distancia genética de estos grupos respecto al grupo Silvestres fue significativamente mayor, presentando una mayor variabilidad genética entre estos grupos.es_ES
dc.description.abstractThe production of the coffee has been traditional in Central America and bound to the socio-economic development, generated 3.8 percent of the gross internal product in 1996, and occupies the third place in the production in the world. The production is concentrated mostly in small farmer hands, furthermore it is an important source of employment generation. The spicie most cultivated in the world is C. arabica originating in the high regions of Ethiopia and Kenya and cultivated in the high lands of Tropical America. The currently cultivated genetic material has as origin a very close genetic base, derived basically from two populations from high freightage Typica and Bourbon as of very few seeds introduced in the century, XVIII and XIX from Holland and the Island of Bourbon. With the discovery and selection of a freightage low growth plant in Brazil, was development the variety Caturra, These one which permits crops easier and highest sowing densities, but susceptible to the coffee rust (Hemileia vastatrix) and the nematodes. Other genetic improvement program was generated as of the Hybrids of Timor, the varieties that derive from the Timor Hybrid are resistant to the coffee rust, to some kinds of nematodes and to some vine-stocks of the antracnosis of the fruit (Colletotrichum kahawae). The objectives of this research were evaluated in the CATIE, Turrialba, Costa Rica, the performance of the molecular markers for the characterization variety of coffee and to evaluate the genetic diversity of the material cultivated (Typica, Bourbon, Wild, and "Introgresados") of coffee by molecular markers and agro-morphological observations. For the agro-morphological characterization were used characteristic of the plant, fruit and grain. It was determined for the morphological characteristics a high genetic variability between varieties and between trees. The variation between varieties is product of the genetic variation, while the variation between trees is product of the influence of the environment, physiological factors and germplasm managing, because the natural homocigosity of the material. The color of the coffee outbreak is a key feature to classify the two basic populations of Typica and Bourbon. The variance components indicate that the factor variety is the one which more contributes to the observed variability, and the heredability index demonstrate that it is feasible to obtain genetic progress in selection programs in the studied varieties. The fruit characteristics, grain and fertility permitted to determine the genetic variability present between groups of coffee. The landfill characteristic of the fruit was presented as the most important to classify varieties between the group Typica and Bourbon. For the molecular characterization was used RAPD. The polymorphism detected was high due to the presence of genotypes of C. canephora and to the group of "introgresados", however, the number of polymorphism markers was under. For middle of the calculation of the similarities between individuals, dendrograms and principal components analysis was estimated the genetic distances between groups, and was generated the classification of the genetic groups. It was identified the different markers between each group and the common markers that are shared between groups.
